ATP Kits

All living cells contain ATP (adenosine triphosphate) and when the cell dies ATP is degraded by intracellular enzymes to ADP and finally AMP. The intracellular ATP concentration is similar in different cell types. Bacterial cells contain 1-2 attomoles per cell while animal and plant cells contain 100 to 10 000 times as much ATP. 

Extracellular ATP may be degraded by adding ATP consuming enzymes or by physical separation. Determination of intracellular ATP can then be used to estimate biomass. If the amount of ATP per cell is known the cell number may also be estimated. 

In sample which likely contains urine, extracellular ATP, ATP in somatic cells and ATP in bacterial cells all three ATP pools can be determined.

ATP kits

Determination of bacterial ATP in urine is used to determine bacteria in the diagnosis of urinary tract infection.
 All ATP assays shall be calibrated by measuring the light before and after adding a known amount of ATP standard at the end of each test. This calibration obviates analytical interference from all sorts of variation of reagents and sample matrices and provides a result expressed in moles rather relative light units.

ATP kit SL

The ATP kit SL is made for monitoring ATP in the range of  10-12 to 10-6 mol/L. The enzymatic formation or degradation of ATP and cellular release of ATP will be monitored by measuring the light intensity. The ATP SL kit is good for 200-1000 assays, depending on the assay volume. The reagents kit contains ATP reagents with luciferase, Diluent, ATP standard and Tris-EDTA buffer.

Microbial ATP Kit

The microbial ATP kit is made for the determination of intracellular ATP in microbial cells, after the elimination of non-microbial ATP. The kit is made for 150-375 assays. The ATP reagents kit contains ATP reagent, with luciferase, diluent, extractant, ATP standard, ATP eliminating reagents and Cell Lysing Reagent.

Intracellular ATP kit

This kit is ideal for determination of intracellular ATP in living cells, as the extracellular ATP will be enzymatic degraded. Subsequently, intracellular ATP is released from the cell by the extractant, which also inactivates the ATP degrading enzyme. With a luminometer the light intensity will be measured before and after addition of the ATP standard. The intracellular kit contains ATP reagents for 150-375 assays. The reagents kit contains ATP reagent containing luciferase and luciferin, diluent, extractant, ATP standard and ATP eliminating reagent. The sensitivity of this kit allows determination of low ATP levels in a variety of circumstances, provided it is not important to degrade ATP in somatic cells.

The Kinase RR kit uses direct bioluminescent measurements of ATP degradation rate. This distinguishes hits from non-hits. Besides that, the standard curve is linear instead of sigmoidal. The Kinase RR kit can be adapted in high throughput screening studies. No radioactive materials, antibodies or other conjugates are needed for these assays.

The Kinase RR kit contains a buffer, ATP standard, ATP reagent, diluent and a blank.
