ATP detection
The core technology of BioThema AB is analytical applications of the luminescent reaction of the Firefly Luciferase enzyme.
We provide luminescence assay reagents for monitoring of the Luciferase enzyme and ATP.
The standard in luminescence technology
BioThema has a unique ATP Standard that is stabilized in liquid to ensure accuracy of each luminometer reading. Calibration with internal Standard ATP accounts for variations between luminometers, inhibition or quenching of light from the sample and sensitivity of reagents.
ATP assay with ATP Reagent HS and Internal ATP Standard
- Add 0.1 ml extractant B/S to a cuvette.
- Add 0.1 ml sample to the cuvette.
- Add 0.4 ml reconstituted ATP Reagent HS and measure light emission, Ismp.
- Add 0.01 ml 10-7 mol/l ATP Standard, i.e. 1 pmol ATP, and measure light emission, Ismp+std.
Calculations
Calculate amount of ATP (pmol) in the sample by the equation:
ATPsmp = 1 * Ismp / ( Ismp + std - Ismp)
The factor 1 is the amount of ATP Standard added (i.e. 1 pmol).
Notes
- The higher the total assay volume the higher the sensitivity of the assay and the more accurate the results
- The (Ismp + std - Ismp) value should be at least 10x higher than the Ismp value. If not, the sample should be diluted or a more concentrated ATP standard used
